“The algorithm-based approaches that people are taking are suboptimal,” says Drew Hannaman, head of R&D at Ichor Medical Systems, which obtained an empirical neoantigen selection platform after Genocea’s bankruptcy last year. “[Algorithm-based approaches] are pretty good at identifying a property that is necessary to generate immune response. But they are not comprehensive enough to identify the properties that are necessary and sufficient.” The result, Hannaman says, is “a lot of false positives”.
"Mass death has long been normalized on Europe’s borders. More than 27,800 people have died or disappeared in the Mediterranean Sea since 2014 — and that is most likely a large underestimate. This year is shaping up to be especially deadly."

"(...) the sole surviving manuscript of the Ethics was deposited in the archives of the Roman Inquisition by Steno, who also submitted an indictment of his former friend to the Inquisitors. They did their work well: their subsequent investigation provides one of the main sources for Spinoza’s life. "

Mary Edwards Walker worked as a surgeon for the Union Army during the Civil War. She was captured by Confederates after crossing enemy lines to treat wounded civilians & arrested as a spy. She's the only woman to ever receive the Medal of Honor - pictured here, wearing it.

'Many late 19th-century paintings present the cypress as the morbid symbol that Hugo describes. Arnold Böcklin’s cypresses in his well-known “Island of the Dead,” for instance, leave little to the imagination. Gabriel-Albert Aurier interpreted van Gogh’s trees in dialogue with these morbid Symbolist paintings.'

@lymphomation It's a strength of the T cell mRNA vaccination approaches coming up that they target multiple epitopes, for Moderna, over 30 per patient.
@lymphomation Yes, absolutely! But unfortunately even there the recurrence rate is still over 50% and a lot of it is due to loss of target antigen or epitope.
I wrote a bit about bispecifics that are coming out (including a cool story involving Francis Crick) that are a potential improvement:

'For example, loss of function of the microglial gene Trem2 renders microglia dysfunctional and causes an early-onset neurodegenerative syndrome, and Trem2 variants are among the strongest genetic risk factors for AD. Thus, restoring microglial function represents a rational therapeutic approach. Here, we show that systemic hematopoietic cell transplantation followed by enhancement of microglia replacement restores microglial function in a Trem2 mutant mouse model of AD.'

@cyrilpedia @halama_immuno @victoralab thank you looping me into this thread.
Fascinating the ipsilateral vs contralateral #vaccination route though perhaps striking thing to me is the lack of a more pronounced signal. I. E. Yes there is a bit to gain from ipsilateral boosting but not a lot.
However I wonder if this could be a crucial variable in achieving effective / longer lasting #immunisation in those hard to vaccinate - e.g. Immunosuppressed
'Both ipsilateral and contralateral vaccination induce a strong immune response, but secondary boosting is more pronounced when choosing vaccine administration-routes that allows for drainage by the same lymph nodes used for priming.'

@lymphomation I agree - in general I am hopeful but very skeptical about any approach targeting a single antigen.
